Dear expert, We unpack and repack MabselectSure LX after 180 cycles. The problem showed after HETP test, the reduced plate height is more than 3 (not as per GE's recommendation) but the asymmetry factor is within specification (0.8-1.8). How to overcome with the high reduced plate height (around 8) and low plate number (around 330). Appreciate your feedback. Thanks

Hi,
there can be several reasons for this low efficiency. If we assume that the packing were performed according to normal procedures then the most common after many purification cycles are that the resin is not cleaned sufficiently (insufficient CIP) to settle in normal manner. Even though the asymmetry are within approved range the efficiency might not be. Also bed support clogging gives much lower plate number as well. Both resin particles and protein residues can cause bed support clogging. In order to give more specific recommendations, more details are required. Please contact your local Scientific support if you need further support.
