Hi experts, I would like to ask during polishing of antibody using CEX process (SP Sepharose) , does the high concentration of samples (10-20mg/ml) will give effects during loading and also elution steps since the sample is too viscous? Is there any study have been done to see the effect of high concentration of samples towards binding capacity of the resins. Appreciate your feedback. Regards, Jehan

We do not have data available for concentrations between 10-20 mg/ml. As you pointed out the viscosity might influence the mass transport into the pores in the chromatography beads which then affect the capacity of the resin. Furthermore, the high protein concentration increases the risk for precipitation when the salt concentration is increased in the elution step. We have tested up to 4 mg/ml and we did not see any changes in capacities of the resin but above that we cannot guarantee that it would be the same capacity.
