Hi, I am a naive user of protein-small molecule studies on SPR. I am planning to check a Protein-small molecule interaction study on Biacore 3000(this is the only available instrument at present to me ). My small molecule is only soluble in 100% DMSO, but further dilutions can be made using milliQ water. I can manage to maintain at the lowest concentration of 0.02% in final interaction step(as analyte). My choice of the buffer is PBS pH 7.4, but as far some of the literature I checked saying that 10X PBS pH will differ from the 1X PBS when we supplement with different % of DMSO. So can I get pH-value at different DMSO concentrations for 10X PBS preparation, especially my DMSO working concentrations are 0.01% to 1%. Your solutions and suggestions are highly appreciated.

Hello,
I have attached a link to our laboratoy guideline "Buffer and sample preparation for direct binding assay in 2% DMSO". It shows in Figure 1, pH as a function of DMSO concentration (Buffer and sample preparation for direct binding assay in 2% DMSO)

Even I am not able to deduce right/exact pH values from that plots for my required 10X PBS pH 7.4

Hi,
0.1-1 % DMSO, is a very low concentration and we do not correct for the refractive index with solvent correction etc. I doubt that there will be any pH change in the buffer. We do not have any data of pH changes with this low concentrations so the easiest way is to check pH if it is affected.
